Ivonescimab plus chemotherapy shows improved overall survival vs durvalumab in first-line biliary tract cancer

In the Phase III HARMONi-GI1 trial, ivonescimab plus chemotherapy met its primary overall survival endpoint with a median OS of 14.9 months versus 12.1 months for durvalumab plus chemotherapy (HR=0.75, 95% CI: 0.61–0.92, P=0.0031), representing a 25% reduction in risk of death. The trial also met key secondary endpoints of PFS and ORR.

Why it matters. Ivonescimab plus chemotherapy could become a new first-line standard of care for advanced biliary tract cancer, offering a statistically significant and clinically meaningful survival benefit over the current durvalumab-based standard.
